Product Overview

Introduction of Janumet 50/1000mg Tablet 56s

Janumet 50/1000mg Tablet is a combination prescription medicine that belongs to the class of oral antidiabetic agents, specifically combining a DPP-4 inhibitor with a biguanide in a single convenient tablet. It contains two active ingredients — Sitagliptin (50mg) and Metformin Hydrochloride (1000mg) — working together in one formulation. Like most medicines, Janumet may cause some side effects in certain individuals, including nausea, diarrhoea, stomach upset, and in rare cases, a serious condition called lactic acidosis, so it is important to take it exactly as your doctor has prescribed. Before you start taking this tablet, it is good to know that this medicine requires a valid prescription, should be taken with meals to reduce stomach discomfort, and is not suitable for everyone — especially those with kidney or liver problems. If you have any concerns or questions about whether this medicine is right for you, always speak to your doctor or pharmacist first.

How to Take Janumet 50/1000mg Tablet

Janumet 50/1000mg Tablet should be taken orally with meals — either with breakfast or dinner — to help reduce the risk of gastrointestinal side effects such as nausea or stomach upset. Swallow the tablet whole with a glass of water and do not crush, split, or chew it unless your doctor or pharmacist has specifically advised otherwise. Always follow your doctor's prescribed dosing schedule and never adjust your dose on your own without professional medical guidance.

Dosage of Janumet 50/1000mg Tablet

The typical recommended dose of Janumet 50/1000mg Tablet is one tablet taken twice daily with meals, providing a total daily dose of Sitagliptin 100mg and Metformin 2000mg, though your doctor may adjust this based on your individual health condition, kidney function, and blood sugar control needs. Never exceed the prescribed dose, and always consult your healthcare provider before making any changes to your dosing routine.

Shelf Life of Janumet 50/1000mg Tablet

Janumet 50/1000mg Tablet generally has a shelf life of 24 to 36 months from the date of manufacture, as indicated on the packaging. Always check the expiry date printed on the blister pack or carton before taking the tablet, and do not use the medicine if it has passed its expiry date, even if the tablet appears unchanged in appearance. Expired medicines should be disposed of safely and responsibly — do not flush them down the drain or throw them in regular household waste.

How to Store Janumet 50/1000mg Tablet

  • Store Janumet 50/1000mg Tablet at room temperature, below 25°C
  • Keep the tablets in a dry place, away from direct sunlight, heat, and moisture
  • Do not store in the bathroom, near a kitchen sink, or in any area with high humidity
  • Keep the medicine in its original packaging until it is time to take it
  • Store out of reach and sight of children and pets at all times
  • If you are unsure about proper storage conditions, consult your pharmacist for guidance

Active Ingredient of Janumet 50/1000mg Tablet

Each film-coated tablet of Janumet 50/1000mg contains the following active pharmaceutical ingredients:

Active IngredientStrength Per Tablet
Sitagliptin (as Sitagliptin Phosphate Monohydrate)50 mg
Metformin Hydrochloride1000 mg

Sitagliptin is a dipeptidyl peptidase-4 (DPP-4) inhibitor, while Metformin Hydrochloride is a biguanide — both are well-established classes of oral antidiabetic agents used in the management of type 2 diabetes mellitus. The combination of these two active ingredients in a single tablet is designed to offer a more convenient dosing option for patients who require dual therapy.

Side Effects of Janumet 50/1000mg Tablet

Like all medicines, Janumet 50/1000mg Tablet may cause side effects in some people. Not everyone will experience them, but it is important to be aware of the following:

Common Side Effects:

  • Nausea and vomiting
  • Diarrhoea or loose stools
  • Stomach pain or abdominal discomfort
  • Flatulence (gas) and bloating
  • Headache
  • Upper respiratory tract infection (e.g., runny nose, sore throat)
  • Decreased appetite

Less Common Side Effects:

  • Hypoglycaemia (low blood sugar), especially when combined with other antidiabetic medicines
  • Dizziness or lightheadedness
  • Constipation
  • Metallic taste in the mouth (associated with Metformin)
  • Skin rash or itching

Serious Side Effects — Seek Immediate Medical Attention:

  • Lactic acidosis — a rare but serious condition associated with Metformin; symptoms include muscle pain, weakness, difficulty breathing, stomach discomfort, nausea, vomiting, feeling cold, or dizziness
  • Pancreatitis — severe and persistent abdominal pain that may radiate to the back, with or without vomiting
  • Severe allergic reactions — including swelling of the face, lips, tongue or throat, difficulty swallowing or breathing, and severe skin rash (Stevens-Johnson syndrome)
  • Kidney problems — reduced urination, swelling in legs or feet
  • Bullous pemphigoid — blistering of the skin, which has been reported with DPP-4 inhibitors including Sitagliptin

⚠️ This is not a complete list of side effects. If you notice any unusual symptoms while taking Janumet, stop taking the medicine and consult your doctor or pharmacist immediately.

Contraindications of Janumet 50/1000mg Tablet

  • Hypersensitivity or allergic reaction to Sitagliptin, Metformin Hydrochloride, or any of the excipients in the formulation
  • Renal impairment or kidney disease — particularly moderate to severe kidney dysfunction (eGFR below 45 mL/min/1.73m²), as Metformin can accumulate and increase the risk of lactic acidosis
  • Hepatic impairment — severe liver disease or abnormal liver function tests
  • Diabetic ketoacidosis or diabetic pre-coma — requires insulin therapy, not oral antidiabetics
  • Type 1 Diabetes Mellitus — Janumet is not indicated for type 1 diabetes
  • Acute or chronic conditions that may cause dehydration — such as severe vomiting, diarrhoea, fever, or reduced fluid intake, which can impair kidney function
  • Acute or recent myocardial infarction (heart attack), respiratory failure, or severe infection that may affect kidney or liver function
  • Excessive alcohol consumption — alcohol increases the risk of lactic acidosis with Metformin
  • Iodinated contrast media procedures — Metformin should be temporarily discontinued before and after procedures involving iodinated contrast agents due to risk of acute kidney injury
  • Pregnancy and breastfeeding — safety has not been fully established; consult your doctor before use
  • Paediatric use — not recommended for children under 18 years of age

⚠️ Always inform your doctor of your full medical history, current medications, and any allergies before starting Janumet.

Drug & Food Interaction of Janumet 50/1000mg Tablet

Drug Interactions:

  • Insulin and other antidiabetic agents (e.g., sulphonylureas) — concurrent use may increase the risk of hypoglycaemia; dose adjustment may be required
  • Diuretics (water tablets) — particularly loop diuretics, may impair kidney function and increase the risk of lactic acidosis with Metformin
  • ACE inhibitors and ARBs — may affect kidney function when used alongside Metformin
  • Corticosteroids — may raise blood glucose levels, potentially reducing the effectiveness of Janumet
  • Beta-blockers — may mask the symptoms of hypoglycaemia such as rapid heartbeat
  • Cimetidine — may increase Metformin plasma levels by reducing its renal excretion
  • Iodinated contrast agents — as mentioned, Metformin should be paused before radiological procedures using contrast dye
  • Certain antibiotics and antifungals — may interact with the metabolism of Sitagliptin; always inform your doctor of any ongoing antibiotic treatment
  • Digoxin — Sitagliptin may slightly increase digoxin plasma levels; monitoring is advisable

Food & Beverage Interactions:

  • Alcohol — avoid or limit alcohol consumption while taking Janumet, as it significantly increases the risk of lactic acidosis and can also cause unpredictable changes in blood sugar levels
  • High-carbohydrate or high-sugar foods — while not a direct drug-food interaction, consuming large amounts of sugary or high-glycaemic foods may reduce the effectiveness of blood sugar management
  • Grapefruit juice — not a major documented interaction with Janumet specifically, but generally advisable to discuss with your pharmacist if you consume it regularly

💡 Always inform your doctor or pharmacist about all medicines, vitamins, herbal supplements, and health products you are currently taking to avoid potential interactions.

Quick Tips for Janumet 50/1000mg Tablet

  • ✅ Always take Janumet with food — taking it with a meal significantly reduces the chance of nausea and stomach discomfort
  • ✅ Never skip doses — consistency is key; if you miss a dose, take it as soon as you remember, but skip it if it is almost time for your next dose — never double up
  • ✅ Keep your follow-up appointments — regular blood tests to monitor your kidney function, liver function, and blood sugar levels are important while on this medication
  • ✅ Store properly — keep tablets below 25°C, away from heat, moisture, and direct sunlight; do not store in the bathroom

  • ✅ Do not share your medication — Janumet is prescribed specifically for you based on your individual health profile; sharing it with others can be dangerous
  • ✅ Carry a medical ID or inform those around you that you are taking an antidiabetic medicine, especially if you are at risk of hypoglycaemia
  • ✅ Inform your doctor before any surgery or medical procedure — Metformin may need to be temporarily stopped before certain procedures, particularly those involving contrast dye
  • ✅ Avoid excessive alcohol — it raises the risk of lactic acidosis, a rare but serious side effect of Metformin
  • ✅ Purchase only from licensed and verified pharmacies — ensure you are buying genuine, registered medication from a reputable source to protect your health and safety
  • ✅ Do not stop taking Janumet suddenly without consulting your doctor, even if you feel well — blood sugar management requires continuous and consistent treatment
