Overview

Agrylin 500mcg Capsule contains anagrelide hydrochloride 500mcg, a platelet-lowering medicine used to treat essential thrombocythaemia, a blood disorder in which the bone marrow produces too many platelets, increasing the risk of abnormal blood clotting or bleeding. It is prescribed and monitored by a haematology specialist.

Anagrelide reduces platelet count by interfering with the maturation of megakaryocytes, the bone marrow cells that produce platelets, lowering the number of new platelets released into the bloodstream over time.

How it Works

Anagrelide selectively inhibits the maturation of megakaryocytes into platelet-producing cells at a late stage of their development, without significantly affecting red or white blood cell production. This gradually reduces the platelet count over 7-14 days of treatment.

Directions for Use

Take Agrylin 500mcg Capsule exactly as prescribed by your haematology specialist, at the same times each day. Swallow the capsule whole with water, and attend all scheduled blood tests and cardiac monitoring appointments, as your dose depends on close follow-up.

Dosage

The starting dose of Agrylin 500mcg Capsule is usually 0.5mg (500mcg) once or twice daily, individualised and titrated by your haematology specialist based on regular platelet count monitoring, with careful cardiac assessment before and during treatment.

Shelf Life

Use this product before the expiry date stated on the packaging. Do not use the medicine if the expiry date has passed.

Storage Instructions

Store below 30°C in a dry place, away from direct sunlight and out of reach of children.

Side Effects

  • Headache
  • Dizziness
  • Nausea
  • Diarrhoea
  • Fast or irregular heartbeat
  • Shortness of breath
  • Fluid retention or swelling (especially in the ankles or legs)
  • Fatigue

Contraindications

  • Known hypersensitivity to anagrelide
  • Severe hepatic or renal impairment
  • Significant pre-existing heart disease, unless carefully assessed and monitored by a specialist
  • Pregnancy, unless the benefit clearly outweighs the risk as determined by your specialist

Drug / Food Interactions

  • Other medicines that lower platelet count or affect bone marrow function may have additive effects requiring closer monitoring
  • Aspirin or NSAIDs combined with anagrelide can increase the risk of bleeding, especially as platelet counts fall
  • Sucralfate may reduce the absorption of anagrelide if taken at the same time
  • Strong CYP1A2 inhibitors (e.g. fluvoxamine) may increase anagrelide blood levels

Quick Tips

  • Attend every scheduled blood test and cardiac check, as your dose is adjusted based on these results
  • Report palpitations, chest pain, or shortness of breath to your haematology team immediately
  • Avoid taking aspirin or NSAIDs without checking with your doctor first, due to increased bleeding risk
  • Never adjust your own dose without your haematology specialist’s guidance

Frequently Asked Questions

What is Agrylin 500mcg Capsule used for?

Agrylin 500mcg Capsule is used to lower an abnormally high platelet count in essential thrombocythaemia, reducing the risk of abnormal blood clotting or bleeding, under haematology specialist supervision.

Why do I need heart monitoring while taking this medicine?

Anagrelide can affect heart rhythm and function in some patients, so your specialist will assess your heart before starting treatment and monitor it periodically throughout, especially if you notice palpitations or chest symptoms.

How long does it take for my platelet count to come down?

Platelet counts typically begin to fall within about 7-14 days of starting treatment, though your specialist will confirm your individual response through blood tests.

Can I take aspirin while on this medicine?

Combining aspirin or other NSAIDs with anagrelide can increase bleeding risk, particularly as your platelet count falls. Always check with your haematology specialist before taking any new medicine, including over-the-counter products.

What should I do if I miss a dose of Agrylin 500mcg Capsule?

Contact your haematology specialist for guidance if you miss a dose, rather than adjusting your own schedule, since your dose is carefully individualised based on monitoring.
