Overview

Revolade 25 mg Tablet contains eltrombopag 25mg, an oral thrombopoietin receptor agonist used to raise low platelet counts (thrombocytopenia) in conditions such as chronic immune thrombocytopenia (ITP), thrombocytopenia associated with chronic hepatitis C, and severe aplastic anaemia that has not responded adequately to other treatments.

Eltrombopag works by binding to and activating the thrombopoietin receptor on bone marrow cells, mimicking the action of the body’s natural thrombopoietin hormone. This stimulates the growth and maturation of megakaryocytes, the bone marrow cells that produce platelets, leading to an increase in circulating platelet count.

How it Works

Eltrombopag binds to the transmembrane domain of the thrombopoietin receptor (TPO-R, also called MPL) on megakaryocyte progenitor cells in the bone marrow, activating signalling pathways that stimulate proliferation and differentiation of these cells into mature, platelet-producing megakaryocytes. This increases platelet production without directly affecting platelet aggregation.

Directions for Use

Take Revolade 25 mg Tablet on an empty stomach, at least 2 hours before or 4 hours after any food or supplement containing calcium or other polyvalent cations (see interactions below), as these significantly reduce its absorption. Swallow the tablet whole with a full glass of water, at about the same time each day.

Dosage

The starting dose of Revolade 25 mg Tablet is usually 50mg once daily (or 25mg in some patients, such as those of East Asian ancestry or with liver impairment), and your doctor will adjust it every 2 weeks based on your platelet count response, up to a maximum of 75mg daily. Treatment requires regular blood tests, and your doctor will individualise both the dose and the duration of treatment for your condition.

  • Statins (e.g. rosuvastatin) may reach higher blood levels when combined with eltrombopag, so your doctor may adjust the statin dose
  • Other medicines that are substrates of certain liver transport proteins (OATP1B1, BCRP) may interact – inform your doctor of all medicines you take
