Overview

Moduride Amiloride HCI 5mg + HCT 50mg Tablet contains amiloride hydrochloride 5mg and hydrochlorothiazide 50mg, a fixed-dose combination of a potassium-sparing diuretic and a thiazide diuretic used to treat high blood pressure (hypertension) and fluid retention (oedema).

Hydrochlorothiazide increases the excretion of sodium, chloride, and water by blocking their reabsorption in the kidney’s distal tubule, lowering blood pressure and reducing fluid build-up, but this can also cause the kidneys to lose potassium. Amiloride blocks sodium channels further along the same tubule, which produces an additional mild diuretic effect while helping the body retain potassium, offsetting the potassium loss that hydrochlorothiazide alone would cause.

Hydrochlorothiazide inhibits the sodium-chloride cotransporter in the distal convoluted tubule of the kidney, reducing sodium and water reabsorption and increasing urine output, which lowers blood pressure and reduces oedema. Amiloride blocks epithelial sodium channels further downstream in the collecting duct, adding a mild diuretic effect while reducing potassium excretion, balancing the potassium-wasting effect of the thiazide component.
