Prograf 1mg Capsule contains tacrolimus 1mg, a calcineurin-inhibitor immunosuppressant medicine. It is used, under transplant specialist supervision, to prevent the body’s immune system from rejecting a transplanted organ (such as a kidney, liver, or heart), and is sometimes used to treat rejection that has not responded to other treatments. Tacrolimus is usually combined with other immunosuppressive medicines as part of a specialist-directed regimen.
Tacrolimus binds to a protein called FKBP-12 inside immune cells; the resulting complex blocks the enzyme calcineurin, which normally activates T-lymphocytes (a type of white blood cell central to organ rejection). By inhibiting calcineurin, tacrolimus reduces T-cell activation and the production of interleukin-2, dampening the immune response that would otherwise attack the transplanted organ.
By forming a complex with FKBP-12 that inhibits calcineurin, tacrolimus prevents the activation and proliferation of T-lymphocytes and reduces production of interleukin-2 and other cytokines. This suppresses the immune response that would otherwise recognise and attack a transplanted organ as foreign tissue.
Take Prograf 1mg Capsule consistently, generally on an empty stomach (at least 1 hour before or 2-3 hours after a meal) at the same times each day, as instructed by your transplant team, since food can affect how much is absorbed. Swallow the capsule whole with water. Do not switch between brands or generic versions of tacrolimus without your doctor’s explicit instruction, as tacrolimus has a narrow therapeutic index and different products may not be interchangeable at the same dose.
