Progyluton Tablet

Progyluton Tablet is a sequential combined hormone replacement therapy (HRT) used to relieve moderate to severe menopausal symptoms such as hot flashes, night sweats, and vaginal dryness caused by declining natural oestrogen levels after menopause. Each pack contains two types of tablets taken in sequence over a 21-day cycle: white tablets containing estradiol valerate alone for the first phase, followed by light brown tablets containing estradiol valerate combined with norgestrel, a progestogen, for the second phase.

This sequential regimen mimics a natural hormonal cycle, allowing the added progestogen to protect the lining of the womb (endometrium) from the effects of unopposed oestrogen. Progyluton Tablet works by replacing the oestrogen that the ovaries stop producing after menopause. Estradiol valerate is converted in the body to estradiol, the main naturally occurring oestrogen, which helps relieve vasomotor symptoms like hot flushes and sweating, and supports vaginal and urinary tract tissue health. Norgestrel, added during the second phase of each cycle, opposes the stimulating effect of oestrogen on the endometrium, reducing the risk of endometrial overgrowth in women who still have a womb.

Progyluton Tablet must only be used under the direction of a doctor following an online or in-clinic consultation with DoctorOnCall. Tablets are taken once daily at approximately the same time each day, in the order printed on the calendar pack, starting with the white (oestrogen-only) tablets followed by the light brown (combined) tablets. After completing all 21 tablets, a 7-day tablet-free interval is observed before starting the next pack, during which a withdrawal bleed similar to a period usually occurs.

The typical regimen is one tablet daily for 21 consecutive days (11 white tablets followed by 10 light brown tablets), followed by a 7-day break with no tablets, then the cycle is repeated. Your doctor will determine the most appropriate starting point and duration of treatment based on your symptoms, medical history, and response, and treatment is generally reviewed periodically to confirm the lowest effective dose is being used for the shortest necessary duration.

Progyluton Tablet is typically stable for up to the expiry date printed on the pack when stored under the manufacturer’s recommended conditions and left unopened. Do not use the medicine after the expiry date has passed.

Store Progyluton Tablet below 30°C in a dry place, away from direct sunlight and out of reach of children. Keep the tablets in their original calendar pack until they are due to be taken so the correct sequence is followed.

• Known, past, or suspected breast cancer
• Known or suspected oestrogen-dependent cancer, such as endometrial cancer
• Undiagnosed abnormal vaginal bleeding
• Current or past history of venous thromboembolism (deep vein thrombosis or pulmonary embolism)
• Active or recent arterial thromboembolic disease, such as stroke or heart attack
• Severe liver disease or liver tumour
• Pregnancy or suspected pregnancy
• Known hypersensitivity to estradiol, norgestrel, or any tablet ingredient

• Enzyme-inducing medicines such as rifampicin, phenytoin, carbamazepine, and St John’s Wort may reduce the effectiveness of Progyluton Tablet
• Certain antibiotics and antifungals may alter hormone levels when taken together
• May affect the required dose of thyroid replacement therapy
• Can influence blood clotting parameters in patients taking anticoagulants
• Grapefruit juice may increase estradiol levels in some individuals

• Take each tablet in the exact order printed on the calendar pack to maintain the correct hormone sequence
• Take your tablet at the same time every day to help you remember and maintain steady hormone levels
• Attend scheduled follow-up reviews with your doctor to reassess whether continued HRT is appropriate
• Report any unexpected vaginal bleeding, breast changes, or leg swelling to your doctor promptly
• Do not share this medicine with anyone else, even if they have similar menopausal symptoms
