Overview
Salofalk 500mg Suppository contains mesalazine (5-aminosalicylic acid, 5-ASA) 500mg, an anti-inflammatory medicine formulated as a suppository for the treatment of ulcerative colitis and, for the suppository form, proctitis. It works directly at the site of inflammation in the bowel lining.
Mesalazine reduces inflammation in the lining of the colon and rectum by blocking the production of inflammatory chemicals (such as prostaglandins and leukotrienes) that drive the tissue damage seen in ulcerative colitis.
How it Works
Mesalazine (5-ASA) acts locally on the cells lining the colon and rectum, inhibiting the cyclo-oxygenase and lipoxygenase pathways that generate inflammatory mediators. This reduces inflammation, ulceration, and associated symptoms such as bloody diarrhoea and abdominal cramping.
Directions for Use
Salofalk 500mg Suppository is inserted rectally, usually after a bowel movement. Wash your hands before and after use. Lie on your side with one knee bent, and gently insert the suppository, pointed end first, into the rectum using your finger, pushing it in far enough that it does not slip out. Try to retain it for as long as possible, ideally overnight, for the medicine to act directly on the lining of the rectum.
Dosage
The usual dose of Salofalk 500mg Suppository for active proctitis (inflammation limited to the rectum) is one suppository (500mg) inserted into the rectum once daily, typically at bedtime. Your doctor will advise on the duration of treatment and may adjust the dose based on your response.

Frequently Asked Questions
What is Salofalk 500mg Suppository used for?
Salofalk 500mg Suppository is used to treat and maintain remission of ulcerative colitis and related bowel inflammation, delivering mesalazine directly to the affected area of the bowel.
How is mesalazine different from sulfasalazine?
Mesalazine is the active anti-inflammatory component of sulfasalazine without the sulfapyridine carrier molecule, which means it generally causes fewer side effects such as rash or blood-related reactions.
How long does it take to see improvement?
Some improvement in symptoms such as bloody diarrhoea and abdominal pain may be seen within 2-4 weeks, but your doctor may continue treatment for several months, including a maintenance phase after remission.
Can I stop this medicine once my symptoms improve?
No, do not stop without consulting your doctor. Continuing mesalazine, even when you feel well, helps maintain remission and reduces the risk of the disease flaring up again.
What should I do if I miss a dose of Salofalk 500mg Suppository?
Take the missed dose as soon as you remember, unless it is almost time for your next dose – in that case, skip the missed dose and continue your regular schedule. Do not double the dose.
